Metrics were mostly inspired by https://crisprdb.org/ go check them and their paper out. Wong N, Liu W, Wang X. WU-CRISPR: characteristics of functional guide RNAs for the CRISPR/Cas9 system. Genome Biol. 2015 Nov 2;16:218. doi: 10.1186/s13059-015-0784-0. PMID: 26521937; PMCID: PMC4629399. I gathered gRNA sequences from them and also from CHOPCHOP for the Homo Sapiens CDKN2A gene. Go check them out as well. Assess and score your gRNA to see if they are suitable for CRISPR deployment. Metrics used: gc_content, good_pos_20, UUU_not_in_seed, no_repetitive_bases, folding_energy, duplex_energy. Disclaimer* The scoring is a bit arbitrary though the metrics themselves should be relatively accurate, feel free to change the scoring to your liking.
